Chimeric antigen receptor T cells (CAR-T cells) refer to T cells that have been genetically engineered to express an artificial T-cell receptor, thereby acquiring anti-tumor capabilities. They are called "chimeric antigen receptors" because the receptor structure simultaneously combines both antigen-binding and T-cell activation functions. CAR-T cell therapy involves several key steps: first, T cells are collected from the patient; next, the CAR construct is delivered into these T cells using a viral vector in vitro; finally, the modified T cells are expanded outside the body before being infused back into the patient. CAR-T cell therapy has already achieved global success in treating B-cell leukemia. Dongling Bio offers CAR lentiviruses targeting the human CD19 molecule, including both human- and mouse-derived CARs, which can be used for in vitro and preclinical research related to CAR-T cell therapies.

Figure 1: Healthy donor-derived CD8+ T cells were infected with Dongling H-41BB-GFP CAR virus. After 72 hours of infection, flow cytometry revealed a CAR-positive T-cell rate of 63% (A). Five days post-infection, CAR-T cell-mediated cytotoxic activity against target cells (CD19+ K562) was assessed, yielding data on the killing efficiency of CAR-T cells at various T-to-target (CD19+ K562) ratios.

Figure 2: CAR-T cells and uninfected CAR control T cells were infused back into NSG mice implanted with Luciferase+RAJI leukemia cells. In vivo imaging was used to monitor the growth of leukemia cells within the mice (A), and survival curves were recorded (B).
