Canine parvovirus
This article discusses Canine Parvovirus (CPV), a highly contagious and often fatal disease that poses significant threats to dogs. CPV is a mandatory pathogen for immunization in the standard-grade viral testing of dogs as per national guidelines, and it must test negative in the nationally recognized SPF viral testing protocol.

Pathology
Canine parvovirus belongs to the genus Parvovirus, family Parvoviridae, and is an enveloped DNA virus with an icosahedral structure. It is characterized by causing vomiting and hemorrhagic enteritis in dogs, specifically due to canine parvovirus type 2 (CPV-2).

Data indicate that among 6,211 VP2 sequences, CPV-2a accounted for the vast majority (45.56%), while the proportion of CPV-2c (29.93%) surpassed that of CPV-2b (22.54%). In contrast, CPV-2/2-like strains—typically originating from vaccine-derived or cross-species-transmitted variants—were relatively rare (1.96%). Prior to 2017, data showed CPV-2a as the dominant variant in Asia, with CPV-2c leading in Europe and Latin America. However, since 2017, the prevalence of CPV-2c has steadily risen, and by 2020, it had overtaken CPV-2a as the new predominant strain. Meanwhile, CPV-2b has remained largely underreported, maintaining a low level of circulation.

Epidemiology
CPV was first isolated in 1978 by American researchers from the feces of dogs suffering from hemorrhagic enteritis. Subsequently, the virus was also detected in most other regions around the world. CPV is highly contagious, typically spreading via the fecal-oral route. Dogs serve as the primary natural hosts for this virus, and animals of all ages and both sexes are susceptible—particularly those aged 4 weeks to 5 years, who exhibit higher susceptibility. Infected dogs are the main source of transmission, shedding the virus extensively through secretions and excretions (infected dogs can continue to excrete the virus in their feces for about 10 days after clinical symptoms appear). Transmission occurs primarily through ingestion, as well as contact with contaminated skin or mucous membranes.
CPV-2 Incidence and Mortality Rates in China
As shown in the table above, while the overall incidence of the virus has decreased since its discovery, the mortality rate among animals has significantly risen, indicating that the virus has become markedly more virulent toward animals.

Clinical symptoms
Canine parvovirus presents with two clinical forms: enteritis and myocarditis. In the enteritis form, the natural infection has an incubation period of 7–14 days and can occur in dogs of all ages (though it’s most common in puppies aged 3–4 months). Early symptoms include fever—often spiking above 40°C—along with lethargy, loss of appetite, vomiting, and diarrhea. Initially, the stool is yellow or gray and quite watery; however, within 2–3 days, bloody stools typically appear. After the onset of blood in the stool, affected dogs rapidly become emaciated, their eyeballs sink inward, and they quickly succumb to severe dehydration and systemic failure, with a disease course lasting about 4 to 5 days and a high mortality rate. The myocarditis form, on the other hand, primarily affects puppies between 4 and 8 weeks of age. Dogs infected with this form often show no obvious clinical signs at first, but suddenly develop severe illness, leading to death within hours. In some cases, puppies may experience mild diarrhea before passing away. Canine parvovirus-induced myocarditis occurs when the virus directly attacks the heart muscle cells, rapidly destroying them and causing symptoms such as labored breathing, cyanosis of the oral mucosa and skin, and, in some instances, sudden collapse and death. This is often the only symptom observed in young puppies suffering from this severe cardiac involvement. Puppies that survive the initial infection, however, may still face long-term consequences due to permanent damage to their heart muscle. As a result, even weeks or months after recovery, these animals can remain vulnerable to fatal outcomes, highlighting the persistent threat posed by this devastating disease.
Diagnostic methods
Blood coagulation and coagulation inhibition assays are simple, inexpensive, and provide intuitive, reliable results; however, these methods are susceptible to various extrinsic factors—such as the type and freshness of blood cells—making them less sensitive compared to molecular biology-based detection techniques.
Molecular biology testing (PCR): Highly sensitive and specific, but the equipment is expensive, time-consuming, and requires operators with advanced technical skills.
Immunological testing (ELISA): While it can detect antibodies in serum, it cannot identify acute infections. Therefore, ELISA is commonly used to analyze antibody levels after vaccination and to help develop immunization schedules for dogs.
Immunoassay Chromatography (ICT): Fast, cost-effective, and simple, delivering accurate and reliable results—but it has lower sensitivity compared to the PCR method.

Prevention and Removal
Canine parvovirus disease has no specific treatment; currently, only symptomatic supportive therapy is available. The mortality rate is high. Infected dogs should be promptly isolated, and the kennel must be thoroughly disinfected and left empty for one month. Dead or dying dogs should be incinerated or buried properly. From a preventive standpoint, timely vaccination remains the fundamental measure to control this disease. Regularly disinfect cages and the surrounding environment using methods such as formalin, β-propiolactone, ultraviolet light, 2–4% caustic soda solution, or 10–20% bleaching powder. Additionally, strengthen overall farm management by maintaining strict hygiene practices, providing nutritionally balanced feed at consistent times and in appropriate amounts.

Research Impact
According to reports, CPV can modulate the body's immune response to other pathogens. CPV also induces significant leukopenia, tumor regression in experimental studies, cerebral white matter softening secondary to myocardial damage, cytokine production and release, rapid blood clotting, and an increased rate of apoptosis.
CPV infection is the leading cause of death from infectious diseases among both wild and domestic dogs worldwide. Despite intensive vaccination efforts, eradicating the virus remains impossible. Yet, dogs are humanity's loyal companions—so for dogs, CPV is essentially a terminal illness. Conquering CPV would be akin to overcoming cancer in humans. As a result, this virus will indefinitely remain a critical concern for professionals working with small animals.
